    Generalized scaling-up approach based on Buckingham theorem for Polymer Electrolyte Membrane Fuel Cells impedance simulation

    Abstract The present paper describes a generalized scaling-up methodology applied to Polymer Electrolyte Membrane Fuel Cells. The use of proper scaling-up algorithms can reduce testing costs within fuel cell manufacturing process by evaluating full stack performance (i.e., impedance behavior) from a single cell/short stack measurement. The algorithm here described relies on a former approach developed by the authors and consists in a generalized methodology combining information measured on single cell and simple physical models (e.g., charge transfer resistance expressed through Tafel equation). A robust technique for the identification of cell reference operational state, such as membrane hydration, from non-scaled data is also introduced. Connection between charge transfer resistance and limiting current is established through diffusion losses modelling. Single cell internal states are estimated by means of inverse models function of numerical intercepts of measured cell spectrum. Stack impedance estimation is then performed through stack internal states assumptions. To prove the consistency and robustness of the proposed methodology, literature data used to design and test the former algorithm version are here considered for algorithm testing and verification

    Temporal repeatability of behaviour in a lizard: implications for behavioural syndrome studies

    It is well established that, across taxa, individuals within populations exhibit consistent differences in their behaviour across time and/or contexts. Further, the functional coupling of traits may result in the formation of a behavioural syndrome. Despite extensive evidence on the existence of consistent among-individual differences in behaviour and behavioural syndromes in the animal realm, these findings are predominately based upon short-term assessments, leading to questions regarding their stability over longer periods. Understanding if these estimates are temporally stable would allow predictions of individual behaviour to be made using short-term repeated measures. Here, we used 57 adult male delicate skinks (Lampropholis delicata) to evaluate the stability of behavioural variation observed both among (animal personality and behavioural plasticity) and within individuals (behavioural predictability), as well as behavioural syndromes, across short (four weeks) and long (five months) timeframes. To do so, we repeatedly assayed activity, exploration, and boldness five times per each individual. Overall, our study revealed complex patterns of behavioural variation and trait (co)variation over time. Activity was always repeatable across time intervals, whereas behavioural differences among individuals in exploration and boldness were not consistent. Yet a behavioural syndrome between activity and exploration was detected at both shorter and longer temporal scales, suggesting that syndrome structure in these traits does not vary as a function of time. Our findings indicate that, at least for some traits (e.g. activity) and studies, short-term measures may be adequate in serving as a proxy for long-term variation in individual behaviour, and to reveal the existence of behavioural syndromes at the population level

    Comparison of different sets of immunological tests to identify treatable immunodeficiencies in adult bronchiectasis patients

European Respiratory Society guidelines recommend a minimum bundle of tests. Broadening the spectrum of immunological tests could increase the number of patients diagnosed with an immunodeficiency and those who could receive specific therapy. The primary objective of the present study was to assess the performance of different sets of immunological tests in diagnosing any, primary, secondary or treatable immunodeficiencies in adults with bronchiectasis. Methods An observational, cross-sectional study was conducted at the Bronchiectasis Program of the Policlinico University Hospital in Milan, Italy, from September 2016 to June 2019. Adult outpatients with a clinical and radiological diagnosis of bronchiectasis underwent the same immunological screening during the first visit when clinically stable consisting of: complete blood count; immunoglobulin (Ig) subclass tests for IgA, IgG, IgM and IgG; total IgE; lymphocyte subsets; and HIV antibodies. The primary endpoint was the prevalence of patients with any immunodeficiencies using five different sets of immunological tests. Results A total of 401 bronchiectasis patients underwent the immunological screening. A significantly different prevalence of bronchiectasis patients diagnosed with any, primary or secondary immunodeficiencies was found across different bundles. 44.6% of bronchiectasis patients had a diagnosis of immunodeficiency when IgG subclasses and lymphocyte subsets were added to the minimum bundle suggested by the guidelines. Conclusion A four-fold increase in the diagnosis of immunodeficiencies can be found in adults with bronchiectasis when IgG subclasses and lymphocyte subsets are added to the bundle of tests recommended by guidelines

    Oleuropein aglycone stabilizes the monomeric \u3b1-synuclein and favours the growth of non-toxic aggregates

    \u3b1-synuclein plays a key role in the pathogenesis of Parkinson's disease (PD); its deposits are found as amyloid fibrils in Lewy bodies and Lewy neurites, the histopathological hallmarks of PD. Amyloid fibrillation is a progressive polymerization path starting from peptide/protein misfolding and proceeding through the transient growth of oligomeric intermediates widely considered as the most toxic species. Consequently, a promising approach of intervention against PD might be preventing \u3b1-synuclein build-up, misfolding and aggregation. A possible strategy involves the use of small molecules able to slow down the aggregation process or to alter oligomer conformation favouring the growth of non-pathogenic species. Here, we show that oleuropein aglycone (OleA), the main olive oil polyphenol, exhibits anti-amyloidogenic power in vitro by interacting with, and stabilizing, \u3b1-synuclein monomers thus hampering the growth of on-pathway oligomers and favouring the growth of stable and harmless aggregates with no tendency to evolve into other cytotoxic amyloids. We investigated the molecular basis of such interference by both biophysical techniques and limited proteolysis; aggregate morphology was monitored by electron microscopy. We also found that OleA reduces the cytotoxicity of \u3b1-synuclein aggregates by hindering their binding to cell membrane components and preventing the resulting oxidative damage to cells

    Lay Summary Prozac is present in freshwater systems across the globe as the pharmaceutical contaminant fluoxetine. The effect of fluoxetine on aquatic species' behavioral variability is not yet clear. We show that male guppies become more similar to each other after exposure to fluoxetine, and females become less flexible in their behavior. These sex-specific differences in response to fluoxetine can have a meaningful impact on their ability to survive in a changing world.The global rise of pharmaceutical contaminants in the aquatic environment poses a serious threat to ecological and evolutionary processes. Studies have traditionally focused on the collateral (average) effects of psychoactive pollutants on ecologically relevant behaviors of wildlife, often neglecting effects among and within individuals, and whether they differ between males and females. We tested whether psychoactive pollutants have sex-specific effects on behavioral individuality and plasticity in guppies (Poecilia reticulata), a freshwater species that inhabits contaminated waterways in the wild. Fish were exposed to fluoxetine (Prozac) for 2 years across multiple generations before their activity and stress-related behavior were repeatedly assayed. Using a Bayesian statistical approach that partitions the effects among and within individuals, we found that males-but not females-in fluoxetine-exposed populations differed less from each other in their behavior (lower behavioral individuality) than unexposed males. In sharp contrast, effects on behavioral plasticity were observed in females-but not in males-whereby exposure to even low levels of fluoxetine resulted in a substantial decrease (activity) and increase (freezing behavior) in the behavioral plasticity of females. Our evidence reveals that psychoactive pollution has sex-specific effects on the individual behavior of fish, suggesting that males and females might not be equally vulnerable to global pollutants

    Psychoactive pollution suppresses individual differences in fish behaviour

    Environmental contamination by pharmaceuticals is global, substantially altering crucial behaviours in animals and impacting on their reproduction and survival. A key question is whether the consequences of these pollutants extend beyond mean behavioural changes, restraining differences in behaviour between individuals. In a controlled, two-year, multigenerational experiment with independent mesocosm populations, we exposed guppies (Poecilia reticulata) to environmentally realistic levels of the ubiquitous pollutant fluoxetine (Prozac). Fish (unexposed: n = 59, low fluoxetine: n = 57, high fluoxetine: n = 58) were repeatedly assayed on four separate occasions for activity and risk-taking behaviour. Fluoxetine homogenized individuals' activity, with individual variation in populations exposed to even low concentrations falling to less than half that in unexposed populations. To understand the proximate mechanism underlying these changes, we tested the relative contribution of variation within and between individuals to the overall decline in individual variation. We found strong evidence that fluoxetine erodes variation in activity between but not within individuals, revealing the hidden consequences of a ubiquitous contaminant on phenotypic variation in fish—likely to impair adaptive potential to environmental change

    Assessing the physicochemical stability and intracellular trafficking of mRNA-based COVID-19 vaccines

    : The emergence of SARS-CoV-2 in Wuhan, China in 2019 has had a profound impact on humanity in every facet. While vaccines against this viral pathogen have been approved a year later, limitations to this therapeutic intervention persist, such as drug sensitivity to transportation and storage conditions, as well as significant financial losses from non-injected resuspended vials. Our research delves into the effects of thermal denaturation (4 - 40 °C) and light irradiation (720 and 10460 kJ/m2) on the mRNA-based vaccines BNT162b2 from BioNTech/Pfizer and mRNA-1273 from Moderna. We also investigated vaccine stability following incubation in syringes to simulate potential interactions with silicon oil. By assaying the effects of these stressors via biochemical and biophysical methods, we aim to elucidate the physicochemical properties, integrity, and stability of these mRNA-based vaccines. Furthermore, the incorporation of a fluorophore into both vaccines allowed us to monitor their localization within cells and assess their capacity to evade vesicular transport mechanisms, thus evaluating the differences between the two formulations. A comprehensive understanding of the aforementioned attributes can enable the establishment of optimal storage and manipulation conditions for these vaccines, thereby ensuring their safe and efficacious application while minimizing the waste of functional and safe therapeutic agents
